Karl von Zierotin was born circa 1500 in Holy Roman Empire to Johann von Zierotin (c.1479-1530) and Anna von Ludanitz and died 20 September 1560 Kolín, Kolín District, Bohemia, Holy Roman Empire of unspecified causes.

Karl von Zierotin was governor (landeshauptmann) of the Duchy of Głogów, imperial advisor and head of the imperial storehouse.

He started his career in the army, fighting for King Louis II of Hungary and afterwards for emperor Charles V, participating in the conquest of Tunis and in the Battle of Mühlberg.

He also supported emperor Ferdinand II who gave him ownership of the Daleschitz monastery. Towards the middle of the 16th century, Karl von Zierotin sold all his estates in Moravia and purchased the estate of Schehuschitz in Bohemia. For his military achievements in Transylvania, the emperor granted him ownership of the village of Kolín and, in 1555 also of the Kolín castle.
