Samuel Wilbore was born in 1594 in Essex, England, to Nicholas Wilbore (1560-1611) and Mary Plumb (1566-1594, their 5th and last child. He married on January 13, 1619 at the age of 24 to the 29 year-old Ann Bradford (1589-1645), who was the daughter of William Bradford (1559-1591), and therefor an older sister of Plymouth colony governor William Bradford (1590-1657).
Samuel and Ann had 7 children:

- Joseph Wilbore (1629-1691)
- William Wilbore (1630-1710)
- Shadrach Wilbore (1631-1697)
- Sarah Wilbor (1633-)
On July 24, 1656, Samuel died in Boston at the age of 62.
